If it is a usb 3.1 of 3.2 drive and port it will most likely run just ok, wouldn’t try it with a 2.0 drive.

Like most games, the files required are loaded into ram. This is what happens in load screens. RAM has high read/write speeds. I’ve noticed textures lagging in GTA V but the game itself remains smooth playing (on ps4) and this has a sata connection. USB 3 is slower than SATA and USB 2 is much slower but the game is much older, only one way to find out!
