Sort of.... A workaround is to install the Voice support App for Amazon devices made by the Dev of SmartTubeNext (named Amazon_SYTV_Bridge.apk).

You can't use the AndroidTV bridge app because it conflicts with the internal YouTube system app in AndroidTV, it even conflicts with the YouTube app when it's disabled.
After the made for Amazon bridge app is installed you can press assistant/search button then ask to "search YouTube for {particular video}" -- this will display an overlay with the YouTube search results and also the SmartTube search results, if it only shows the YouTube results then exit and search again; not perfect, but it does work most the time.
Because of Google Play Services being integrated to everything in AndroidTV it will always display "Video" search results that point to YouTube first, even if that app is not even installed, there is no way possible for Google Assistant to "only" search through SmartTubeNext and not YouTube when both apps are searching the same domains.
No. That app's developer would have to build the basic ability to be recognized by Assistant.
Go ask the developers of it to add support.
